Sabouraud Dextrose Medium

Sabouraud Dextrose (SabDex), formulated by Sabouraud, is used for culturing dermatophytes13 where the pH is adjusted to approximately 5.6 in order to enhance the growth of fungi, especially dermatophytes, and to slightly inhibit bacterial growth in clinical specimens2. This medium is recommended for mold and yeast counts by the U.S. Pharmacopeia, Standard Methods for the Examination of Water and Wastewater, the Association of Official Analytical Chemists, and the Compendium of Methods for the Microbiological Examination of Foods3,6,14-16. Sabouraud Dextrose Broth is a modification of the original formulation without agar and is recommended by the U.S. Pharmacopeia for sterility testing of pharmaceutical products14. Sabouraud Dextrose Agar with Lecithin and Tween 80 is used to neutralize the antimicrobial effects of disinfectants or cleaning solutions used on environmental surfaces when cultivating fungi from these surfaces.
Chloramphenicol and/or tetracycline and/or gentamicin may be added as broad spectrum antimicrobials to inhibit the growth of a wide range of gram-positive and gram-negative bacteria.

User Quality Control



Check for signs of contamination and deterioration. Users of commercially prepared media may be required to perform quality control testing with at least one known organism to demonstrate growth or a positive reaction; and at least one organism to demonstrate inhibition or a negative reaction (where applicable).

Following are the composition for SabDex derivative media: SabDex Broth is the same formulation as above, without agar added (Final pH 5.6 +/- 0.2 at 25°C). SabDex Agar with Chloramphenicol contains 50.0 mg of chloramphenicol (Final pH 5.6 +/- 0.3 at 25°C). SabDex Agar with Chloramphenicol and Gentamicin contains 50.0 mg of chloramphenicol and 5.0 mg gentamicin. Final pH of 5.6 +/- 0.3 at 25 degrees C. SabDex Agar with Chloramphenicol and Tetracycline contains 50.0 mg of chloramphenicol and 10.0 mg of tetracycline. Final pH of 5.6 +/- 0.3 at 25 degrees C. SabDex Agar, Emmons has only 20.0 g of dextrose. Final pH of 6.9 +/- 0.2 at 25 degrees C. SabDex Agar with Lecithin and Tween® 80 contains 0.7 g of lecithin and 5.0 gm of Tween® 80.
